No we did not use different colors for different sex puppies. The first puppy born was automatically #1 and we charted all the info immediately such as time of birth, birth weight, sex of puppy and which toenail we painted and so on for the next puppy born. There were 2 of us during the whelp which made it easier. We never had any confusion and all the information was charted perfectly!
