Are her nipples pink and larger than usual? By 5 weeks you can usually notice a change in them - also you might notice a slight thickening in the belly area, but if there is only one puppy it is sometimes longer than 5 weeks before you notice a change.
Regarding organogenesis, I'm not sure what you can do to avoid damage. I wouldn't give Heartguard or Frontline if I think she is pregnant, even though they say it is safe. I wouldn't take the chance.
