I have personally never had a smear performed, but I know you can detect progesterone levels with a blood test and this is usually used in situations with artificial insemination. Most likely you would have to have the test performed several times. Only some labs are capable of giving results in 24 hours, most send them off and it takes 3 days which defeats the purpose.
Most of the time the male and female instinctively know the proper timing. In your situation I would use a proven stud and just get the pair together starting on Day 10 everyday until they tie and then every other day until she will no longer accept mating. I have heard of dogs not ovulating until like 25 days or more after the onset of the heat so every dog is different.
