I remember when Thumper had his first 3 year rabies shot after I rescued him his back thigh was sore for a day or two. He is usually a very docile dog but he ended up trying to bite me anytime I got near him. I took him back to the vet that day because I was so concerned (new dog mom at the time and didn't know what was going on)

The vet assured me he was just tender and it would go away. It did after a day. However, it was scary going through it and watching my baby suffer for the day.

I don't like to give pain meds unless necessary. So unless my baby was in extreme pain I would wait it out. Maybe a cold compress around the site or if allowed some type of tylenol would help.
