Yes a Cysto is done in the office. It's very easy for the Vet to do. They take a needle and draw the urine directly from the bladder. It does not hurt the dog at the all. This is actually the most sterile way to get a urine sample.

I have had it done several times on mine.

If when the Vet feels the bladder and it doesn't feel full, they will ask for you to leave your dog there and they will keep checking the bladder for fullness.
As for expense I think my Vet charged me $20.00.
