If Jessie never needed her glands manually expressed before, I wouldn't do it again (after you clear up this problem). I agree with Deanna about having your vet take a look.
When Max was a puppy, he scooted and had gland problems after a vet retrieved a stool sample manually. He had to be manually expressed a couple more times and then never again. He is 4.5 years old now and expresses naturally.
