Bach flower is a stress reliever and I know a lot of people who use it on their dogs and it works very well.
Sounds like you need to start right at the beginning with introducing your dog to the car.
First just let her sit with you on the back seat with no engine on just let her get used to the smell of the car etc, make it fun give her a treat.
Do this a few times and gradually increase the time.
Then let her sit in the car with the engine running but don't go anywhere so that she get's used to the engine noise.
Do you have her in a crate in the car? As this makes them feel more secure.
Just try very short journey's at first and reward her with a treat when you let her out.
Make lot's of short car journey's with a lovely walk when you get there, that way she will quickly learn that a car ride isn't just assosiated with a visit to the vet but there maybe a pleasurable experience at the end of the journey.
Give her lot's and lot's of praise.
I would try this before the tranquiliser route.
Also try the bach flower remedy.
Hope this helps
