Your pup does need a rabies vaccine by law if you are in the US. Some very bad things can happen if it is not given (legally).
However, IMO it is best not to do it on spay day. Most dogs do just fine when this is done, but they are already going through so much. No reason to add extra stress when it isn't necessary. And with all the other drugs that will be given, if there is a reaction you won't know what caused it.
