Is the rug itself backed with some of that rubbery non-slip stuff? I ask because for some reason, that rubbery stuff attracts both dogs and cats to potty on it. I have had that problem with a couple of bath mats before and had a salesperson tell me not to get that type but instead get one that is just fabric only, without the rubber backing. I switched out all of my bathmats and kitchen rugs and never had that problem again. If your rug isnt like that, you still might consider swapping rugs to see if that helps.
